Information about the derived orientation of disease risk-increasing variants provides power to detect signals of directional selection on diseases
Tests for directional selection on disease-associated variation typically use effect sizes and allele frequencies. This project shows that polarizing risk-increasing alleles by their derived or ancestral state carries additional information, and builds that orientation into a more powerful test for directional selection on disease phenotypes.
